Suitabi­lity assessment of microbiological purity testing procedures was performed. It has been established that the studied­ product in standard testing methods possessed antimicrobial activity against test microorganisms Basillus subtilis ATCC 6633 (on soybean casein digest agar and Sabouraud dextrose agar containing an antibiotic), Candida albicans АТСС 10231 (on soybean casein digest agar and Sabouraud dextrose agar containing an antibiotic), and possessed no antimicrobial activity against test microorganisms Staphylococcus aureus ATCC 6538 (on soybean casein digest agar) and Pseudomonas aeruginosa ATCC 9027 (on soybean casein digest agar). The efficacy of lecithin and polysorbate as substances eliminating antibacterial effect in combination with 50-fold dilution of the test sample has been suggested and proven for the purpose of antimicrobial activity neutralization. One mushroom that has shown potent medicinal properties both by oral consumption and topical application is the Tremella fuciformis . GC/MS analyses of T. fuciformis oil obtained by hydro-distillation showed four predominant compounds. 9,19-Cyclolanost-24-en-3-ol, (3.beta.)- was the most predominant with R T of 20.766 and 24.672 min and a percentage total of 32.681 followed by 7-Isopropenyl-1,4a-dimethyl-4,4a,5 ,6,7,8-hexahydro-3H-naphthalen-2-one with R T of 22.425 min and a percentage total of 21.334, lupeol had R T of 22.825 and 23.230 min and a percentage total of 18.531, while lanosterol had R T of 21.079 and 23.034 min and a percentage total of 17.845. These four compounds represent 90.391% of the total constituent compounds in T. fuciformis oil. Horbachevsky Ternopil State Medical University, Ukraine; e-mail: hnkafor@yahoo.com In this research, we investigated the effect of pituitrin-izadrin induced injury on the levels of metallothionein (MT) and soluble and filament forms of glial fibrillary acidic protein (GFAP) in the hippocampus, cerebellum, thalamus, and the cerebral cortex, and examined the effect of corvitin on the brain under the noted changes. Our results showed oppositely directed changes – a decrease in the level of MT and an increase in GFAP in the rat brain, with a tendency to astrogliosis development, under the influence of systemic deficiencies in myocardial function. Mykhalojko Ivano-Frankivsk National Medical University, Ukraine; e-mail: myhalojko@i.ua The aim was to determine the alterations in levels of vasculair endothelial growth factor (VEGF), homocysteine (HC) and lipid profile in patients with acute ischemic stroke (AIS), to establish the relationship between them. A clinical and neurological examination of 120 patients aged 56 to 75 years with AIS was done. Diagnosis was established taking into account results of spiral computer and magnetic resonance tomography. The control group included 20 people with no signs of acute cerebrovascular and severe somatic pathology aged 59.35±1.8 years. Assessment of disability was performed at admission and during the treatment using the scale NIHSS. In all relevant patients in 1-2 days after stroke, the concentration of HC and VEGF levels was determined by ELISA. Blood lipid profile was determined by enzymatic cоlorimetric method. The study found that increasing concentration of plasma HC is associated with intensity of hyperlipidemia, and hyperhomocysteinemia severity prevails over the changes in the lipid profile components. During AIS a significant increase in the level of VEGF in the blood was observed, indicating the presence of vascular accident and activation of angiogenesis. We aimed to investigate the capacity of MSC obtained from young mice to modulate arginine metabolism of macrophages from old animals. Bone marrow cells obtained from young and aged mice were cocultivated with MSC in the presence of M-CSF. Nitric oxide production was analyzed in supernatants by Griess reaction, and arginase activity was measured in cell lysates. We have found that arginase activity was significantly lower in macrophages isolated from old mice as compared to young animals ( P ˂ 0.05). Syngeneic MSC addition markedly stimulated arginase activity in macrophages from both young and aged mice ( P ˂ 0.001), with greater effect in old animals. There were no significant differences in nitric oxide level between groups. But the mechanisms of such regulation are still discussed. In this paper we have shown that Ca ions directly or indirectly regulate the level of myometrium mitochondria membranes polarization. The additions of 100 µM Ca 2+ were accompanied by depolarization of the mitochondria membranes. The following experiments were designed to study the impact of Ca 2+ on the myometrium mitochondria [Ca 2+ ] m . Isolated myometrium mitochondria were preincubated without or with 10 μM Са 2+ followed by 100 μM Са 2+ addition. Experiments were conducted in three mediums: without ATP and Mg 2+ (0-medium), in the presence of 3 mM Mg 2+ (Mg-medium) and 3 mM Mg 2+ + 3 mM ATP (Mg,ATP-medium). It was shown that the effects of 10 μM Са 2+ addition were different in different mediums, namely in 0- and Mg-medium the [Ca 2+ ] m values increased, whereas in Mg,ATP-medium statistically reliable changes were not registered. Preincubation of mitochondria with 10 μM Са 2+ did not affect the [Ca 2+ ] m value after the addition of 100 μM Са2+. The [Ca 2+ ] m values after 100 μM Са 2+ addition were the same in 0- and Mg,ATP-mediums and somewhat lower in Mg-medium. Preliminary incubation of mitochondria with 10 μM Са 2+ in 0- and Mg-mediums reduced changes of Fluo 4 normalized fluorescence values that were induced by 100 μM Са 2+ additions, but in Mg,ATP-medium such differences were not recorded. It is concluded that Са 2+ exchange in myometrium mitochondria is regulated by the concentration of Ca ions as in the external medium, so in the matrix of mitochondria. The medium composition had a significant impact on the [Са 2+ ] m values in the absence of exogenous cation. Since the lithium ion radius is close to that of magnesium, lithium ion can compete with magnesium as cofactor of certain magnesium-dependent enzymes. Numerous studies have shown lithium to be an inhibitor of glycogen synthase kinase 3. It also inhibits the phosphatases involved in the metabolism of phosphoinositides and phosphoadenylate 3′-nucleotidase. Iutynska Zabolotny Institute of Microbiology and Virology, National Academy of Sciences of Ukraine, Kyiv; e-mail: bilyuvskal@gmail.com, galyna.iutynska@gmail.com The goal of this work was to study biosynthesis of sterols by soil streptomycetes Streptomyces avermitilis IMV Ac-5015, Streptomyces netropsis IMV Ac-5025 and Streptomyces violaceus IMV Ac-5027 on various media under submerged cultivation and to determine their content in bioformulations, developed on the basis of these metabolites producers. Analysis of sterol derivatives extracted from biomass, supernatant of cultural liquid and biological products were performed by GC/MS. The biomass of streptomycetes contained sterols in significantly higher amounts than in the cultural supernatants, and their spectrum and quantitative ratio were different. Squalene – sterol precursor – was found in the biomass in quantities producers 6.2-43.3 μg/g, when streptomycetes grown in synthetic and 8.2-212.1 μg/g in organic media. The biomass of S. avermitilis IMV Ac-5015 was distinguished by domination of 24-epibrassinolide, the content of which was the highest among the tested strains and reached 268.2 μg/g in organic and 345 μg/g in synthetic media. S. avermitilis IMV Ac-5015 did not synthesize sitosterol and stigmasterol that is important in respect of nematicidal properties of the strain. The biomass of S. netropsis IMV Ac-5025 and S. violaceus IMV Ac-5027 grown on organic medium contained 2.3-6.8-fold higher amount of sitosterol and 1.5-3-fold higher amount of stigmasterol compared to synthetic medium. Ergosterol prevailed in sterol spectrum of S. netropsis IMV Ac-5025 and S. violaceus IMV Ac-5027. The highest total content of sterols (9.5 mg/l) was found in Avercom (producer – S. avermitilis ІМV Ас-5015).
